I have tubing on both of those ports.
You could do a test. Raise the tubes above the carbs & blow into them one at a time. The one that's open is the overflow. For the other one, unscrew the drain screw several turns & blow again. If it's now open, then that one would be the fuel level check (or carb drain).

Intresting, I've never seen 2 ports. My pumpers on my '80 F1 have only the one port at each carb.
