The FSM shows a manual petcock.
If the petcock at hand is vacuum operated, assure its vac hose is connected to the carb's vacuum spigot.
The remaining two carb spigots are probably for vent hoses exiting to open air.

A couple of things about your carbs that are different than the carbs on my 79'E/Shafty.
Your carbs don't have the accelerator pump/fuel bowls anymore. (also, the outboard bowls seem out of position as the fuel drains aren't facing outwards).
The vacuum ports are (from L to R in your photo) #2 vent, #3 fuel petcock, #4 vent.
