Will a fouled plug clean itself with use?

I messed up an over oiled my pod filters, whilst fitting new plugs.,,and other things. I broke a golden rule about not messing with ignition and carbs at the same time so It took me a while to work out what I'd done wrong.

The new plugs are now sooted, will normal use clean them up over time -assuming my mixture not too far out now?

How long does it take to put a deposit on a plug enough to read it? I've read 1 mile WOT, others 10 seconds, preferably uphill.

The other thing being, if most of my riding is 1/4 to 1/2 throttle, will a plug chop at this speed be a good indication?

I don't see why not. Plugs are designed to clean themselves. You can do plug chops cold, then hot to compare.
