Well that dia, did you use a drill for a gauge? Is that exact? From what I see that is larger than the #32 Drill listed in the Dyno Jet kit, which is .1155".
Yes a hole in the Diaphragm is bad, it will cause the slide to rise slower than the others, less fuel from the needle jet circuit, and probably a hesitation on that cylinder.

it was most likely a .1155 but it drilled over.

I recently checked my jets and they installed 140 and 160's does that sound about right for the bike I have with mac 4 into 1 with pod filters

if u have a hole in the diaphram, u will want to get that fixed before u adjust anything else.
