Idle adjustment knob

Hello forum,
My bike is 1982 kz550ltdc. I'm in the process of checking floats, with the clear tube method. Before this is done,
my concern is the idle adjustment knob. Must this be set at a certain position? I'm not sure if it must be turned a certain way, or just backed all the way out. Any input from the forum would be greatly appreciated!

Idle thumbscrew position is irrevelant when dealing with fuel level in floatbowls.
