OK than I will than presume that these are KEIHN carburetors and not Mikuni. They are the only Goofy company that puts the lift holes at the intake side of the slide. In this case, I believe that they should both be drilled out. The drill they give you is only a .098". On the Mikuni carbs for the same bike they give you a .116 drill. this is for a single hole.

Ok so to comfirm, I will drill out both holes with the drill they gave me.

Yes, because the holes are evenly spaced off the centerline

My Mikuni BS34's have two holes in bottom of slider, one is for the needle and the other I assume is an air hole. Is it the air hole that needs to be drilled??

Yes the air hole is what gets drilled. It is the one that is off-center. The center hole is for the needle.
But you don't necessarily have to drill the hole. Enlarging it just makes the slide jumpier. That is. yes it will go up faster, and if the engine can handle it, it might make for a bit quicker acceleration, but it also makes the slide more sensitive and "bouncier", meaning it moves up and down during engine pulses during intermediate throttle positions.
If the bike is having trouble running, then I would not drill the slide as a remedy. I don't think it would ever be the problem. Many people never drill the slide so they can go back to stock. It just means the slide opens a tiny bit slower than necessary.

I'm just a little confused as to what it will take for my 1982 GPz 1100 w/ 4 into 1 V&H megaphone (no baffel) and Pod filters to smooth out that first 1/8th throttle, ...I read somewhere on here, ... shim stock needles .040" to .050", ..along with 40 pilots and 140 main jets. Ethan over @ MikeXS said to put a shim P/N SU009-395 under the needles and it should solve the issue.
Just to clarify, does shim go under the needle's clip (tapered end pointing down), then into slider??
