If they are between the carb and the intake manifold, it's a vacuum port. If it's between the carb and the air filter, it's a bowl vent.

they look like stock vm28's to me. some had accel. pumps, some didn't. those came on late 70's kz's. as far as 4 individual ports? don't know!! i have a set and they only have 2...

I had to order a set of those plugs for a set of carbs I rebuilt. Z1 Enterprises carries them.

In your first pic, the passage on the outside of the carb (where the gas passage would have been) looks like it might be plugged with epoxy. It looks like your set may have been made from combining 2 sets of carbs with 2 X #2 position carb and 2 X #3 position carbs. That would explain the many ports on your carbs.
They should work fine, but make sure that all the bowls have some sort of vent. Plugged bowl vents can cause all sorts of erattic running. And make sure that the vents that have vacuum when running are plugged or ran to your vacuum petcock.

On the left side of Conti's carb, below the idle mixture screw, where the gas passage would be located.
If you look at Rons carb, the same spot is open (not filled- yes, they are pumper carbs but the later VM series look like Rons on the outside).
It looks like an inner carb body may have replaced the outer left carb body. If this passage wasnt filled, the carbs would leak gas out of it.
The carb gas passage could have had the other side of the passage drilled (or it may have been open , depending on the application) to get it to work correctly.

Anyway, I cleaned the carbs on today:
I'll replace the needs, gaskets etc on next week-end. The float needle of carb #3 was sized!

I have a 1979 kz1000st also and that is the vacuum that goes to your fuel tank petcock. If this wasn't connected your bike shouldn't have run, unless you have a different petcock.

The carbs should work fine, just make sure all the parts (jets, needles, etc.) are the same on all 4.
Check the slides to be sure they are the same (the cut out size is stamped on the bottom of the slides)
