Do you mean you are having trouble getting the nut loose from the sprocket (assuming that is your setup-you didn't mention what bike), or trouble getting the sprocket itself off the output shaft?
If it's the latter, then you probably have some corrosion in the splines. Any good gear puller should work. If it's getting the nut loose, you have to put about as much torque on the wrench as was used to tighten it. My KZ750 requires 58ftlbs to tighten, and about as much to loosen. Could be that someone put too much torque on it, or used some strong Locktite! Also, remember to loosen the bent-over lock washer (if it uses one) before trying to remove the nut. You could also spray some PB Blaster around the threads and let it set a while before you try it.

Use a hair dryer at the hottest setting to loosen up stuck fasteners. Avoid a torch/flame as you might ignite the oil or grease in the area.
Tapping it with a hammer will also break the rust/corrosion bond loose.

I just use an impact wrench and it comes right off. Those sukas are super tight. Make sure the engine is in neutral and the chain is on otherwise you can damage the gears inside and you will not be able to take it off. The chain must hold the sprocket so it will not rotate, not the transmission.

If you don't have a good impact wrench, then I did the following:
With the bike on the center stand, place a cinder block on its end straight out from the nut. Put a piece of wood on top of the cinder block. Use a 1/2" or larger drive break-over bar, and an extension of about 8" long. Put the head of the break-over bar on top of the wood and have the handle a little higher than horizontal. This will keep it in alignment with the center of the nut.
Have the transmission in neutral and the chain still on the sprockets. Have a helper to press on the rear brake pedal. Now the risky part - stand on the handle end of the wrench. It helps if you have something like a post close by to hold onto. You may need to jump on it to break it loose. With years of tension and heat cycles it may take 150 ft./lbs. to get the nut loose. Good luck.

I agree with Otakar. If you don't have an impact wrench, take it into any repair shop and see if they will take a try at it. I do that and have it retightened enough to get back home and then do what I need to do. Some have charged me a few bucks but most do it as public relations and wish me luck. I go back to those shops for parts and sevice when I need to. Help those that help you!
