No, there should not be any difference, have you measured the primary resistance(small wire side)?
You should see 3-4 ohms, if one is higher that the other, I'd replace both ignition coils.

Testing Kawasaki Ignition Coils For Input Voltage

When voltage testing Kawasaki Ignition Coils, the following needs to be done:

1. Remove the fuel/gas tank to access the coils.

2. Fully charge the battery.

3. Using a multimeter, set it up for VDC(Volts, Direct Current), range of 20.

4. Turn on the ignition switch and the run/stop switch to “Run”.

5. Put the multimeter’s RED probe on where the red or red/yellow wire goes on the ignition coil.

6. Put the multimeter’s BLACK probe on either the battery Negative(-) post/terminal or a good frame ground.

7. You should be able to see battery voltage at the ignition coil connection, but this depends on the current draw of other items in the same circuit.

8. If the tested voltage is down to 8-9 volts, you probably need to clean the many and various electrical connectors.

9. To check the spark output at the sparkplugs, use Ignition Spark Testers. Instead of grounding the sparkplug with the sparkplug wire attached, looking for a spark. This method puts the IC Igniter and ignition coils at risk of failure due to high currents, and possible internal shorting.

These bikes have a 5000 ohm, non-replaceable resistor in the sparkplug caps. They go bad from heat, vibration and age. New ones are available at most Japanese motorcycle stores, or through Z1 Enterprises, Inc. - Specializing in Vintage Japanese Motorcycle Parts, including New Old Stock & aftermarket parts. www.z1enterprises.com is their website.

I believe the 750 Spectre has the same coils as my 81 KZ650-CSR of which the following applies:
The coils are marked (+) & (-) on the primary ends where the igniter leads connect.
The red wires should be connected to (+) . Black & green to (-) .
The black wire goes to the right side (1-4) coil & the green wire goes to the left side (2-3) coil.
The specified primary resistance is 1.8 - 2.7 ohms, but keep in mind that the coils can test fine cold but could be faulty under load. A simple check is to ride the bike at highway speeds, then stop & feel the coils to see if they're getting hot to the touch --- press hard w your fingers (don't get burned by the valve cover). They should be warm but not hot. Mine were getting hot, so I got green Dyna 3.0 ohm coils from Z1Enterprises.com.

Again, the above is for the 81 650-CSR, but I believe your bike has the same ignition system.
If you need a manual, search eebbaayy for KZ750 Shaft Manual.
There are usually quite a few available, with 1 or 2 at reasonable prices.

You could use the dyna grey coils 2.4 ohms... greater spark. That is what I have in my 750 LTD

Per FSM for 1983 kz750, resistance of ignition coil primary winding is 1.8 ~ 2.8Ω.

What are the test results for the coil to 1 and 4?

If needed, may download the FSM with this link:
