When you say "fresh rebuild" does that mean you split the cases?

I'm trying to understand your problem. What I think you wrote is that when you use the kickstart your cam chain does not rotate. But also, when you use the kickstart the crank rotates. Is that accurate?
I have several restored bikes along with a 2006 Goldwing with a sidecar. My wife has a 2019 Suzuki DR 650 for on and off road.

If the cam chain rotates when you use a spanner on the crank ignition side end bolt then the cam chain is on the crank shaft sprocket.
If the cam chain doesn't rotate when using the kick starter then the crank shaft also isn't rotating therefore the kick starter is not engaging.
Can you feel cylinder compression when using the kick starter?

Is the clutch installed ? (I believe the crank will turn independently of the "first motion" shaft until the clutch is assembled)

Ditto Wookies comment, clutch has to have the plates in it or no kicker action. I asked about if you had split the cases because you might of disassembled the ratcheting mechanism on starter shaft? These HAVE to be indexed correctly or the kicker won't work! Or work correctly depending how far off the punch marks are off. Shown the two types, early (hollow shaft) and later solid. But note the punch marks on shaft and rachet stopper. You have to align these marks when installing the rachet stopper. Remember back what you fiddled with??? Unfortunately if incorrect then you will have to split the cases again, uggg.
