New to KZrider so forgive me if this has been a topic before.  I tried searching for previous topics on this but nothing came up.

I am trying to remove the drive sprocket nut but am having troubles.  The Clymers manual states to, “have an assistant hold the rear wheel to keep the drive chain taut”.  Unfortunately the engine is not in the bike.  

Any other tips or tricks for removing it?

Welcome to KZRider. I fixed your pictures to be full size.  Next time click the insert all tab to complete the installation. 
Best to include what bike / motor you are working on  when posting .
   You can use a impact gun to loosen the nut and also wedge the sprocket teeth with a piece of wood being carefull not to alloew the wedge to push against  the rod shown or a edge of the  engine  / trans  casting  ( Just be careful )
 Others will take a old  chain and  secure it over the sprocket  and clamp it so it cant spin  while loosening the nut .

Follow the advice about using a rattle gun, don't try it with a breaker bar - speaking from bitter experience!

If the top end, or at least the bottom end is still together it might help to put the trans in 1st. gear when using an impact gun on that nut.  That'll introduce some additional friction & inertial mass to work against.  If it's in neutral the shaft might just spin.

It doesn't seem like a good idea to me to put the shock loads from an impact gun through the transmission gears.  I really don't think that would be good for gear teeth.  It's not the type of load they were intended to take.  It think using a chain wrench to hold gear seems like a much a better idea.

What size socket does it need? Clymers doesn’t say.  Still working at getting that nut off.

Measure across the flats of the nut. It will tell you what size socket you need

It would be of help to know just which engine/model you are working with. If you have posted this and I did not see/find it then I apologize for that. But from the pics/pages you posted it appears to me to be a (k)Z650, but what do I know. With knowing just which model/year we may be able to provide you with the size of the nut as there are some differences in them over the years/models.

Kawasaki sometimes lists the size. 
For example, for the 1977 KZ650
