I have had my KZ1100 running for about 6 months when I noticed some misfiring and now the no 2 & 4 cylinders aren't firing (slightly warm or cold) header pipes. Compression is between 140-150, all cylinders are getting spark (replaced plugs and wires) and I had done a basic rebuild on the carbs awhile ago (replaced boots as well.. I took the carbs off, there is gas in all the bowls and made sure all the jets are clear. What might I be missing? Next on my list is doing a valve adjustment but I don't think valve adjustment is an issue given I have good compression and this problem surfaced in a short amount of time. Thanks for any advice.

You mentioned the jets, but are all the fuel and air passageways clear as well?

My engine is loving a little more fuel in the mix these cooler days. I bumped up to 20 pilots, and am seriously considering 22.5's.

I don't know the bike very well. Does it have an exciter to boost the pickup signal?

I have several restored bikes along with a 2006 Goldwing with a sidecar. My wife has a 2019 Suzuki DR 650 for on and off road.

It has the typical cdi ignition. Getting strong spark again after replacing all plug wires. I disassembled carbs again and made sure everything was clear and no debris at the bottom of the bowls. Did find that the jet plug o-rings were not sealing. I had a set of used rubber plugs leftover from another project and used those to temporarily fix the problem. If my carburetor knowledge serves me correctly, this would have caused a rich conditions and maybe caused fouling?
Took the opportunity to do a bench sync (one of the adjustment screws had come loose and readjust float level (lowered it a bit). Now it works great! Running stronger than ever, pulling smoothly right up to 8500. Still
